Bonds, Catch Bonds, and Statistics

چکیده
Unsurprisingly, education research shows that students engage better when an instructional storyline begins with a surprising claim about topic important in their own lives. Today, everyone understands the importance of immune response to lives, and most find it paradoxical be told some bonds strengthen under applied pulling force. So recent discovery catch-bond behavior T-cell activation is very good starting point motivate study many molecular biophysics ideas. To understand claim, we must discuss nature bonds, replace “strength” by statistical notion mean lifetime, then introduce random walks on energy landscapes. Rather than elaborate technical Kramers theory, however, can readily perform simple simulations. The results include memorable animated graphics, make themselves platform such as Python or MATLAB, yield conceptual insight into bond formation breakage, isomerization, so on. Abstractions Boltzmann distribution exponentially distributed lifetime emerge concrete consequences rules, origin catch bonding clear multiple unbinding routes are available. Catch bonds
What has been your biggest error? Back at the beginning of this century, I used to say “I am going to spend the rest of my life trapped in a mitochondrion!”. This was because I was convinced that it was never going to be possible to retrieve even partial extinct genomes. Now, there are genomic sequences for many ancient humans and animals. This goes back to the famous Arthur C. Clarke quote: “W...
متن کاملCatch bonds in adhesion.
One of the most exciting discoveries in biological adhesion is the recent and counter-intuitive observation that the lifetimes of some biological adhesive bonds, called catch bonds, are enhanced by tensile mechanical force. At least two types of adhesive proteins have been shown to form catch bonds--blood proteins called selectins and a bacterial protein called FimH. Both mediate shear-enhanced...
متن کاملPolymer-based catch-bonds.
Catch-bonds refer to the counterintuitive notion that the average lifetime of a bond has a maximum at a nonzero applied force. They have been found in several ligand-receptor pairs and their origin is still a topic of debate. Here, we use coarse-grained simulations and kinetic theory to demonstrate that a multimeric protein, with self-interacting domain pairs, can display catch-bond behavior. O...
متن کاملPhenomenological and microscopic theories for catch bonds.
Lifetimes of bound states of protein complexes or biomolecule folded states typically decrease when subject to mechanical force. However, a plethora of biological systems exhibit the counter-intuitive phenomenon of catch bonding, where non-covalent bonds become stronger under externally applied forces. The quest to understand the origin of catch-bond behavior has led to the development of pheno...
متن کاملReceptor–ligand binding: ‘catch’ bonds finally caught
Leukocyte recruitment to sites of inflammation is initiated by the selectin family of adhesion receptors. Recent research reveals that P-selectin binding to its ligand exhibits 'catch' to 'slip' bond transition that may help explain the shear threshold phenomenon.
